The Simoa® Cytokine 4-Plex A (C4PA) Advantage PLUS Assay (Product # 104979) measures IL-1β, IL-6, IL-10, and TNF-α at levels as low as fg/mL, providing reliable quantification of these key inflammatory cytokines. The levels of these 4 cytokines measured in blood offer valuable insights into inflammation and immune responses to various conditions, such as infections, injuries, cancers, and immunotherapy. The fully automated HD-X Analyzer provides scalability of up to 6 plates in an 8-hour day, accomplishing in a single day what manual preparation methods would take a week to complete.

The target cytokines of Simoa® C4PA Advantage PLUS
IL-1β: IL-1β is a potent pro-inflammatory cytokine mainly produced by macrophages. It is a key mediator of immune and inflammatory response to infection and injury, protecting the body against infectious or traumatic insults. Excessive production of IL-1β in chronic inflammation can cause or enhance the manifestation of many diseases such as neuroinflammatory conditions like Alzheimer’s, rheumatoid arthritis, and inflammatory bowel diseases.
IL-6: IL-6 is a broad-acting cytokine that elicit both pro-inflammatory and anti-inflammatory outcomes It is rapidly induced upon tissue damage or inflammation due to infections or injuries, stimulating acute phase immune reactions. IL-6 also plays a key role in adaptive immunity by promoting the differentiation of activated B cells into antibody-producing cells and naïve T cells into effector T cells. Dysregulated IL-6 has been implicated in autoimmune disease, chronic inflammatory disorders and cancers.
IL-10: IL-10 is an anti-inflammatory cytokine that regulates immune responses by inhibiting the production of pro-inflammatory cytokines. It is mainly produced by regulatory T cells and macrophages to prevent excessive immune activation and tissue damage. In line, IL-10 can limit the pathogenesis of inflammatory diseases. Tumor cells can produce IL-10 that may contribute to their ability to cause immune suppression.
TNF-α: TNF-α is a key pro-inflammatory cytokine in systemic inflammation, playing a central role in the regulation of immune cells, inducing fever, apoptosis, and promoting the acute phase immune response. Overproduction of TNF-α can be a pathological component of inflammatory and autoimmune diseases like rheumatoid arthritis, inflammatory bowel disease, and psoriasis. TNF-α can be a therapeutic target to control inflammation in these conditions.

Simoa® C4PA Advantage PLUS is a digital immunoassay for the quantitative determination of IL-1β, IL-6, IL-10, and TNF-α concentrations in human serum and EDTA plasma samples.
